Protein measurement with the Folin phenol reagent

Abstract

The use of the Folin phenol reagent with proteins which have been treated with an alkaline Cu reagent is recommended for the estimation of protein in, e.g., enzyme fractions and antigen-antibody precipitates. The absorptiometric procedure described is suitable for amounts of protein as small as 0.2 µg. Care is necessary in the use of the reagent, since the intensity of colour varies with the protein and is not proportional to the concentration of protein
